Will this language setting affect update?

I have it on a UK male setting for speech and text the regular US setting. Will it affect the update to cortana?

No , this won't , you will get windows phone 8.1 if you are enrolled in dev preview program. You have to change your phone region to enable cortana.

In settings, go to language and region and country and region to US and restart phone, everything will be ok.
