If I install build 10162 (or whatever the latest official ISO build is) and then try to update to a later build, what will be the download size?
Will it download the whole ISO again or will it just download the required patches?

If you update through Windows Update, it'll only download the Updates.
Wait for someone to confirm too.

Insider builds are full downloads and installs at around 3.3GB. When we get the final build, updates will come via Windows Update, and should be small. I expect we'll get one or maybe 2 more insider builds that are full size downloads.

So will I be able to pause and resume downloads as I am on a REALLY slow connection?

Really slow connection user here. You can't, Windows Update doesn't support resuming download if transmission is paused in the middle of file, it will restart for that particular file.
