I was about to tell Cortana who my brother is but accidentally tapped another contact. How shall I change my brother that Cortana knows?

Go to her settings there u'll find inner circle go there click the one you want to change then on right bottom press three dots menu will pop up to see details or delete from inner circle. Delete it and add again
